Analytical Method Transfer — Protocols, Acceptance Criteria & Comparability
Method transfer demonstrates that a receiving laboratory can execute a validated method with equivalent results. It succeeds or fails on the protocol: defined samples, acceptance criteria and a clear record of what was compared and what deviated.
Write the protocol before the experiment
Define the method and version, the samples (including, where possible, the same lots at both sites), the analysts and instruments, and the statistical acceptance criteria. Comparative testing, co-validation and transfer waivers are different strategies; justify the choice. Ambiguous criteria invite retrospective rationalization when results are borderline.
Control the known sources of difference
Instruments, columns, reagents, sample preparation details and analyst technique drive most transfer failures. The sending laboratory should supply the method's critical parameters and robustness knowledge, not only the written procedure. Agree how system-suitability failures and investigations are documented.
Close the transfer with a usable report
The report should state what was compared, the results against each criterion, deviations and the conclusion on transfer success. Where a method will run routinely at the receiving site, define ongoing comparability expectations. Transfer evidence supports the quality system; it does not revalidate the method from scratch unless criteria fail.
Sample considerations
- Provide the method, validation summary, critical parameters and available reference samples.
- State the sending and receiving sites, the transfer strategy and the timelines.
- Agree samples, replicates, acceptance criteria and the investigation plan for failures.
Questions to ask a laboratory
- Which transfer strategy applies, and what are the acceptance criteria?
- What method knowledge beyond the written procedure will be shared?
- How are deviations and failed criteria investigated and documented?
