IVD Specimen, Reagent & Calibrator Stability
Separate specimen handling and collection-container effects from reagent, calibrator and control stability. Define the assay-specific conditions and claims to be supported.
Specimen handling and collection containers
Identify matrix, collection device/additives, separation delay, transport temperatures, storage intervals and freeze-thaw history. Study representative analyte levels and decision thresholds under justified worst-case conditions. Assess container adsorption, contamination or additive interference where plausible; one stable specimen type does not establish stability for every matrix.
Reagents, calibrators and controls
Separate unopened shelf life, transport stress, opened/in-use stability and instrument onboard stability. Define lot strategy, reference assignments, traceability, reconstitution and handling, and performance checks tied to the intended assay. Accelerated observations need appropriate real-time evidence and justified models; do not invent expiry periods or claim a single lot represents every formulation.
Compare proposals by deliverables
Ask for study design, time points, sample and lot allocation, stability-indicating endpoints, predefined acceptance criteria, raw data, deviations and change-control recommendations. Clinical performance requires its own rationale; a stable reagent does not establish clinical sensitivity or specificity.
What to include in your request
Start with the intended decision and available evidence.
- Intended use, research-only versus clinical purpose, biomarker/analyte and decision to support; disclose confidential identities only in private follow-up.
- Assay format, instrument/software versions, qualitative/quantitative output, specimen matrix, collection container, handling and storage conditions.
- Available reference materials, reagent/calibrator lots, candidate/comparator method, target range or decision threshold and existing evidence.
- Proposed claims, report and study design, sample availability, ethics/consent requirements, budget, location and milestones; ask the provider to propose justified methods.
